She had a brief relationship with fellow actor Paul Rudd. Clarke joined Kappa Alpha Theta while she was a student at Indiana University. She was initiated into the Beta part in 1990. Clarke became interested in acting while at the same time studying abroad during her senior year in Bologna, Italy.
When she got back to the United States, she started taking architectural photography classes. She studied acting at Circle in the Square Theatre School, Axis Theater Company, and The Willow Cabin Theatre Company, and she was given free acting lessons in exchange for taking photographs at a cultural arts center.

Clarke made her acting debut in a Volkswagen commercial that won an award in 1999. She followed this up with a role in the 2000 short film Pas de deux, for which she won the Brooklyn Film Festival’s Outstanding Performance award.
With minor roles in films like All About George in 2000 and The Accident in 2001, as well as television shows like Ed and Sex and the City, Clarke’s career quickly took off. Clarke put in an audition for the role of Nina Myers, a CTU agent, on 24 in 2001.
On the day that filming began, she was awarded the role. She had to wear her own outfit for the entire season of filming because the wardrobe department did not have time to fit her. Clarke was in 36 episodes over the course of the show’s three seasons.
Clarke won a Brilliant Satellite Honor for Best Execution by an Entertainer in a Supporting Job in a Show Series for this job. Clarke also provided 24 with her voice: The Game, where she played Nina Myers from 24.
She has appeared as a guest on Life and House. In Twilight, she played Bella Swan’s mother, Renée Dwyer. She accepted the main role in the TNT series Trust Me, opposite Eric McCormack and Thomas Cavanagh.
The series debuted on January 26, 2009, to positive surveys, however, it was dropped after one season due to declining ratings. In 2010, Clarke repeated her role as Renée Dwyer in Eclipse, the third film in the Twilight series. She was given the role of Marla Jameson in the 2013 American version of The Tomorrow People.
